Characterization of Starch Hydrolysis of Amylase from Paenibacillus polymyxa PC-1

Abstract
We screened to obtain amylase-producing microorganism from environmental soil isolated from Sangju-si, Gyeongbuk, Korea. As a result, we selected one strain due to its ability of forming active zone on LB agar plate containing starch. In the analysis of 16S rRNA gene sequence from the strain, the isolated strain showed 99% of identity with Paenibacillus polymyxa, therefore we named the strain Paenibacillus polymyxa PC-1. When P. polymyxa PC-1 was cultivated with LB medium with 0.5% starch, the strain produced amylase maximally after 5 days of cultivation. The amylase from P. polymyxa PC-1 showed the highest activity at pH 6.0 and 50¡É and was stable up to 50¡É and at range of pH 6.0~8.0 against 30 min of treatment. In the hydrolysis of starch, the amylase from P. polymyxa PC-1 exhibited maximum activity in the 2% of starch concentration. And, under the condition (pH 6.0, 50¡É , and 2% starch), the enzyme produced 5.73 mg/ml of reducing sugar after 180 reaction.
